Grissom, the series points leader, lost a lap because of a cut tire on the fourth of 250 laps on the one-mile track at Wisconsin State Fair Park.
But his fast car and racing luck allowed him to make up his lap during a series of green-flag pit stops by the race leaders on lap 178.
During the race's final caution, which flew when Roy Payne crashed in turn 2 on lap 234, Grissom chose to pit for fresh tires while most of the other front-runners remained on the track.
When the race was restarted with nine laps left, Grissom had dropped from seventh to eighth, but quickly got into contention on the new tires.
The lead changed hands twice in the last five laps. Larry Pearson passed David Green to lead laps 246 through 248, then Grissom slipped by Pearson to lead the final two miles.
Grissom, driving a Chevrolet, nipped Pearson's Chevrolet by .56 seconds. It was the Gadsden, Ala., native's second win in 14 races.
Rookie Hermie Sadler finished third in an Oldsmobile, with Green fourth in a Chevrolet and Tom Peck fifth in an Oldsmobile.
The first NASCAR event at the historic Milwaukee track in eight years attracted an estimated 40,000 fans. Local officials are hoping to eventually land a Winston Cup race.
The top finisher from four Winston Cup regulars competing was Darrell Waltrip, who took 12th. Jerry Glanville, coach of the NFL's Atlanta Falcons, caused three of the eight caution flags with spins and twice hit the wall. He finished 26th.
