Berteotti made a 15-foot birdie putt on the 18th hole to subdue Mochrie, who had come from five strokes behind at the start of the final round to force the playoff.
Lynchburg's Donna Andrews, who had a three-stroke lead entering the final round, shot 74 and tied second-day leader Helen Alfredsson of Sweden for fourth at 279. Alfredsson shot 71. Danielle Ammaccapane was third, two strokes off the lead after a 68.
Berteotti, who began the day three strokes off the lead, and Mochrie finished with 12-under-par 276 totals. Mochrie shot a 66 and Berteotti 68 on the 6,137-yard Blue Hill Country Club course.
On the final playoff hole, a par-5, both golfers left their third shots about 15 feet from the cup, with Mochrie about two inches farther away.
She left her uphill putt from in front of the hole about one inch short and swept the club through the air in frustration. She tapped in for a par. Berteotti, to the right of the cup, then rolled it in to grab the winner's check of $67,500. She jumped several times and punched the air with both fists.
Mochrie missed an eight-foot birdie putt that would have won on the first playoff hole. Then, Berteotti saved par on the second playoff hole by one-putting.
On the third playoff hole, Mochrie's 30-foot birdie putt from the back fringe lipped out and she made par. Berteotti then two-putted from 15 feet. On the next playoff hole, Mochrie hit her second shot in a bunker but blasted out and made a par 4.
Berteotti took a share of the lead at 9 under when Andrews bogeyed the third hole. Berteotti went ahead by herself at 10 under with a birdie at the 133-yard seventh.
